A cell-permeable pyrazolopyrimidine compound that acts as a potent and ATP competitive inhibitor of mTOR (IC50 = 4 nM). Active in LNCap cellular assay (IC50 = 355 nM). Selective against PI3K’s (p110a IC50 = 1026 nM, p110b, p110g, p110d > 6000 nM). Selective against other PIKK’s and other kinases. When injected into tumor-bearing nude mice, WYE-354 inhibited mTORC1 and mTORC2 and displayed robust antitumor activity in PTEN-null tumors.
